It can be measured in the serum or plasma by liquid chromatography-mass spectrometry (LC-MS) methods, which are sensitive and require little sample volume. The LC-MS method measures the molecular ions of 4²-hydroxycholesterol, which are then converted to mass spectra using a mass spectrometer. The mass spectrum is then used to identify the formic acid adducts of 4²-hydroxylated cholestanol molecules.
