Vafidemstat (ORY-2001) is a lysine-histone demethylase (LSD1)/MAO-B inhibitor for the study of neurological disorders.

Vafidemstat is a tetracycline compound that has been shown to be effective in the treatment of neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease. It is able to penetrate the blood-brain barrier and act on the central nervous system. Vafidemstat inhibits hyperactivity by increasing the concentration of gamma-aminobutyric acid (GABA) in the brain, which may be due to its ability to inhibit chloride transport into cells. Vafidemstat also increases GABA concentrations in lymphatic vessels, which may contribute to its anti-inflammatory effects.
