Venlafaxine is a selective serotonin and norepinephrine reuptake inhibitor (SNRI) that is used to treat depression. Venlafaxine inhibits the reuptake of serotonin and norepinephrine into the presynaptic neuron, increasing their availability in the synaptic cleft. Venlafaxine has been shown to be effective in treating major depressive disorder and other depressive disorders, such as dysthymia and post-partum depression. The antidepressant effects of venlafaxine are believed to be due to its ability to block the uptake of serotonin and norepinephrine in the brain, resulting in an increased concentration of these neurotransmitters. R-(-)-O-desmethylvenlafaxine is a prodrug of the antidepressant drug venlafaxine. It is converted in vivo to the active form of venlafaxine, which inhibits the serotonin transporter. In this study, R-(-)-O-desmethylvenlafaxine was used to determine whether it has a greater effect on depression than on other symptoms such as hyperactivity in children. A gravimetric analysis showed that there was no difference between the groups for weight gain or changes in heart rate. However, there were significant differences in other parameters such as appetite and sleep patterns, highlighting the need for further research into the effects of this drug on children with depression and attention deficit hyperactivity disorder (ADHD).
