Chlorthalidone-d4 is a chlorthalidone prodrug that is hydrolyzed in the body to release the active form, medoxomil. Chlorthalidone-d4 has been shown to be bioequivalent to medoxomil after oral administration in humans. It has been used in pharmacokinetic studies and has demonstrated a high degree of absorption, rapid clearance, and low protein binding. The drug is metabolized by CYP2C9 and CYP3A4 enzymes into inactive metabolites. Chlorthalidone-d4 also binds to plasma proteins in the blood with high affinity. This drug has been used for the treatment of hypertension or edema of various origins such as congestive heart failure or cirrhosis of the liver.
