    Thymidine-5'-triphosphoric acid (TTP) is a nucleotide analog that is used in the treatment of bowel disease. TTP inhibits bacterial growth by binding to DNA polymerase, thereby preventing transcription and replication. It also interrupts DNA synthesis by inhibiting the incorporation of deoxynucleotides into DNA. The enzyme activity of TTP has been studied using an experimental model in which the bowel disease was induced by administration of dextran sulfate sodium (DSS). X-ray crystal structures have shown that TTP binds to metal chelates and forms intermolecular hydrogen bonds with nucleotides in the major groove of double-stranded DNA. In addition, it has been shown that TTP binds specifically to nuclear DNA and not to mitochondrial DNA or RNA.
