PA452 is a selective antagonist of retinoic X receptor (RXR) and suppresses the effect of Retinoic acid on Th1/Th2 development.

PA 452 is a synthetic analog of erythromycin, which has been shown to inhibit the growth of malignant cells and to be effective in treating metabolic disorders. It is also active against viral life and stimulates the production of epidermal growth factor. PA 452 binds with high affinity to nuclear hormone receptors and inhibits fatty acid synthesis. PA 452 also inhibits the expression of insulin resistance, which may be due to its ability to bind with high affinity to hepatocyte-like cells. This drug has been shown to induce apoptosis in malignant cells that are resistant to all-trans retinoic acid (ATRA) or hydrogen peroxide (H2O2).
