CAS:Virstatin represents a small-molecule inhibitor of Vibrio cholerae virulence and intestinal colonization. By inhibiting the transcriptional regulator ToxT, it prevents expression of two critical V. cholerae virulence factors, cholera toxin and the toxin coregulated pilus. It has been shown to improve insulin sensitivity and reduce inflammation in animal models of inflammatory diseases. 4-(1,3-Dioxo-1H,3H-benzo[de]isoquinolin-2-yl)-butyric acid also binds to the insulin receptor and inhibits mitotic checkpoint proteins in human cells. The binding constants for this molecule have been determined using tryptophan fluorescence spectroscopy on type strain bacteria.
