Tacrolimus, con el número CAS 104987-11-3, es un fármaco inmunosupresor utilizado principalmente para prevenir el rechazo de órganos en pacientes trasplantados y para tratar ciertas condiciones autoinmunitarias. Es una lactona macrólida derivada de la bacteria Streptomyces tsukubaensis. Tacrolimus funciona inhibiendo la activación y proliferación de linfocitos T, principalmente a través de la inhibición de la calcineurina, una enzima crítica en la señalización de células T. Este mecanismo ayuda a suprimir la respuesta inmune. La sustancia se administra típicamente en su forma tópica para condiciones de la piel como la dermatitis atópica o como una formulación oral para uso sistémico. Tacrolimus es lipofílico, lo que influye en su absorción y distribución en el cuerpo. Tiene una vida media relativamente larga, lo que requiere un monitoreo cuidadoso de los niveles en sangre para evitar toxicidad. Los efectos secundarios comunes incluyen un mayor riesgo de infecciones, nefrotoxicidad y neurotoxicidad. Debido a sus potentes efectos inmunosupresores, Tacrolimus a menudo se utiliza en conjunto con otros inmunosupresores para mejorar la eficacia mientras se minimizan los efectos adversos.

    CAS:
    FK-506 is an immunosuppressant that is produced by fermentation. It is used to prevent the rejection of transplanted organs and to treat a number of autoimmune diseases, including rheumatoid arthritis and multiple sclerosis. FK-506 binds to the IL-2 receptor on T-cells and inhibits their proliferation, thereby preventing activation of other immune cells. FK-506 has been shown to be effective in treating a number of diseases, including myocardial infarcts, coronary heart disease, and oral hypoglycaemic. The optimum concentration for this drug is not known, but it appears that higher concentrations are more beneficial in treating some conditions.
