Lumefantrina es un compuesto antipalúdico que se utiliza principalmente en combinación con arteméter para el tratamiento de la malaria, particularmente en áreas con cepas de Plasmodium falciparum resistentes a la cloroquina. Se clasifica como un derivado de metanol de fenantreno y exhibe una estructura química compleja que contribuye a sus propiedades farmacológicas. Lumefantrina se caracteriza por su lipofilia, que mejora su absorción y biodisponibilidad cuando se administra por vía oral. La sustancia tiene una vida media relativamente larga, lo que permite regímenes de dosificación efectivos. Actúa inhibiendo el proceso de detoxificación de hemo en el parásito de la malaria, lo que lleva a la acumulación de hemo tóxico y, en última instancia, a la muerte del parásito. Lumefantrina generalmente es bien tolerado, aunque puede causar efectos secundarios como trastornos gastrointestinales y, en raras ocasiones, problemas cardíacos debido a su potencial para prolongar el intervalo QT. Su eficacia se maximiza cuando se utiliza en terapias combinadas, que ayudan a reducir el riesgo de desarrollo de resistencia en los parásitos de la malaria.
