Descrizione:
Furantoina, noto anche come nitroFurantoina, è un antibiotico utilizzato principalmente per trattare le infezioni del tratto urinario. Appartiene alla classe dei composti nitrofuranici e presenta un'attività antibatterica ad ampio spettro contro vari batteri Gram-positivi e Gram-negativi. La struttura chimica di Furantoina presenta un anello nitrofuranico, essenziale per le sue proprietà antimicrobiche. Agisce inibendo la sintesi delle proteine batteriche, del DNA, dell'RNA e della sintesi della parete cellulare, portando infine alla morte cellulare batterica. Furantoina viene tipicamente somministrato per via orale ed è ben assorbito nel tratto gastrointestinale, con la sua efficacia aumentata se assunto con cibo. Gli effetti collaterali comuni possono includere disturbi gastrointestinali, reazioni allergiche e, in rari casi, tossicità polmonare. A causa del suo specifico meccanismo d'azione e farmacocinetica, Furantoina è particolarmente efficace per le infezioni del tratto urinario non complicate causate da organismi suscettibili. Tuttavia, non è adatto per infezioni sistemiche o quelle causate da alcuni batteri resistenti. Nitrofurantoin's mode of action involves the inhibition of essential bacterial enzymes. It is reduced by bacterial flavoproteins to active intermediates, which subsequently alter or disrupt ribosomal proteins, DNA, and other vital macromolecules within bacterial cells, thereby inhibiting key metabolic processes. Nitrofurantoin is most commonly used in the treatment and prophylaxis of uncomplicated urinary tract infections (UTIs). Its effectiveness is primarily restricted to the urinary tract because it achieves therapeutic concentrations only in urine. It is particularly active against Escherichia coli and some strains of Enterococcus, Klebsiella, and Staphylococcus. Due to its targeted spectrum and localized action, it is generally reserved for infections caused by susceptible organisms within the urinary tract, reducing the risk of systemic side effects.
