Description:Lipid A is a component of the outer membrane of Gram-negative bacteria. Lipid A is a complex molecule that is composed of three major domains: an amino-terminal domain, a lipid tail, and a carboxy-terminal domain. The amino-terminal domain contains two receptor binding sites that are responsible for activating the innate immune system. Peptides in the amino-terminal domain bind to Toll-like receptor 4 (TLR4) and activate macrophages, which release inflammatory cytokines and nitric oxide. The lipid tail interacts with CD14 on macrophages, which leads to the activation of nuclear factor κB (NFκB). Activation of TLR4 and CD14 causes an increase in the production of proinflammatory cytokines.
