Description:Pamapimod is a drug that inhibits the production of TNF-α and IL-1β. It has been shown to reduce the severity of rheumatoid arthritis in humans, as well as to improve the symptoms of other autoimmune diseases such as psoriasis and inflammatory bowel disease. Pamapimod is also active in animal models for these diseases, suggesting that it could have a potential use as an anti-inflammatory agent in the treatment of human autoimmune disorders. This drug binds to TNF receptors on immune cells, preventing them from binding to TNF-α and IL-1β. Pamapimod is able to inhibit both soluble and bound forms of TNF receptor 1 (TNFR1) and TNFR2 with high affinity, but does not inhibit TNFR3.
Pamapimod has been found to be safe at doses up to 10 mg/kg in humans, with no serious adverse events reported at this dose level. In rats, p
