    CAS:
Clofarabine is a nucleoside analog that is used to treat various types of cancer, including leukemia, lymphoma, and solid tumours. Clofarabine has been shown to have minimal toxicity in healthy cells and is usually well tolerated by patients. It acts by inhibiting the synthesis of DNA in rapidly dividing cells. This effect can be synergistic with other anti-cancer agents such as the cytotoxic agent Bleomycin. Clofarabine binds to an intracellular target and inhibits DNA synthesis by blocking the enzyme DNA polymerase. Inhibition of this enzyme leads to cell death. Clofarabine has been tested in pediatric patients from age 1 month old up to 18 years old. The drug has been found to be safe for use in this population.

2'-C-Ethynylcytidine is a synthetic surfactant that can be used in coatings and other surface treatments. It has been shown to inhibit the growth of bacteria and viruses, as well as being effective against a variety of bacterial species. 2'-C-Ethynylcytidine binds to sulfide, which is produced by bacteria, leading to cell death. The antimicrobial activity of 2'-C-Ethynylcytidine is due to its ability to form ester linkages with polyesters, which are able to penetrate the bacterial cell membrane and disrupt the integrity of the cytoplasmic membrane. This leads to leakage of intracellular contents, followed by cell death. 2'-C-Ethynylcytidine also has been shown to bind to metal ions such as silver, copper, zinc and cadmium and prevent their uptake into cells.

Adenosine-5'-triphosphate (ATP) is a nucleotide found in all living cells. It is used as a coenzyme for adenosine kinase, adenosine phosphokinase, and adenosine deaminase. ATP also acts as an important signaling molecule and has been shown to be involved in the regulation of cell metabolism, muscle contraction, and other cellular processes. The human body normally produces enough ATP to meet its needs through aerobic metabolism of glucose or fatty acids, but under certain conditions such as intense muscular activity or insufficient oxygen, more ATP must be produced by anaerobic metabolism. In these situations the mitochondria use glycolysis to produce lactic acid which is then converted into pyruvic acid by the enzyme lactate dehydrogenase. Pyruvic acid is then oxidized by the enzyme pyruvate dehydrogenase complex to produce acetyl-CoA which is further oxidized by the enzyme complex called Krebs

    CAS:
5-Carbamoylmethyluridine is a nucleoside that is structurally related to uridine. It is synthesized from uridine by methyl esterification with carbamic acid and methoxycarbonyl chloride. 5-Carbamoylmethyluridine interacts with actin filaments and has been shown to alter the expression of messenger RNA in eukaryotes. 5-Carbamoylmethyluridine also acts as a competitive inhibitor of the formation of a ternary complex, which includes ATP, GTP, and UTP, at the level of ribonucleotide reductase. This inhibition leads to decreased synthesis of messenger RNA in cells.

It has been shown that pseudothymidine can be incorporated into DNA without disrupting the polymerase chain reaction. The thermal denaturation of pseudothymidine is 10 °C lower than that of thymidine, allowing it to be used at higher temperatures with less risk of degradation. Pseudothymidine may also have medical applications, such as its use in cancer treatment. 8-Methylthioguanosine is a purine nucleoside that is synthesized from guanosine by the enzyme nucleoside phosphorylase. 8-Methylthioguanosine is used in the treatment of leukemia cells and has been shown to have homogeneity, with a single peak on paper chromatography. It is also an inhibitor of purine nucleotide phosphorylase, which prevents the conversion of guanosine to inosine monophosphate and leads to depletion of intracellular ATP levels. This drug binds tightly to the active site of purine nucleotide phosphorylase and inhibits its activity. 8-Methylthioguanosine may be used as a radiosensitizer for cancer therapy and can be administered intravenously or orally.

  • 3'-O-Methyluridine

    CAS:
3'-O-Methyluridine is a nucleoside that is used in the preparation of oligonucleotides and as a substrate for uridine phosphorylase. It is used to study the termination of RNA synthesis, which occurs when there are no more ribose residues in the RNA template strand. When this happens, an adenosine residue is added by polyphosphate kinase to the 3' end of the RNA strand. This addition causes a release of pyrophosphate and leads to end of DNA synthesis. The acid hydrolysis technique can be used to separate 3'-O-methyluridine from other nucleotides. This reaction produces guanosine, which can be detected using polymerase chain reaction (PCR) techniques such as polyacrylamide gel electrophoresis (PAGE).

2'-Deoxy-2'-fluoroadenosine-5'-diphosphate (FAD) is an irreversible enzyme inhibitor that binds to the active site of the decarboxylase enzyme. FAD binds to cytochrome P450 and inhibits aromatase, which is an enzyme responsible for the conversion of testosterone to estradiol. It has been shown that FAD inactivates the enzyme by protonation at the active site. The protonated form of FAD is a nucleophile and forms a covalent bond with the substrate, thereby inhibiting decarboxylases.
