The hydrochloride salt of 4IM, koenigicine, has been shown to produce estrogen levels in the female rat similar to those produced by 17β-estradiol. The synthesis of 4IM involves the nitration of 2-methylanisole followed by iodine oxidation. This reaction produces a mixture of mono-, di-, and tri-iodonapthalenes.
